遠在三百餘年前,即明末清初,在現在嘉義縣朴子市東南方約一公里許,是片雜草蔓生,遍地荒蕪,缺山岳為依,乏大湖為憑,兼有「康榔」遍野,內中有條彎彎曲曲九九彎盤龍似的「大龍溝」,溯源自康熙二十年涂姓祖先等六房十八員,由大陸福建省漳州府詔安縣仙堂荷仔埔社,奉帶天上聖母渡台開墾,自福建漳州來的涂姓祖先暫宿於此,睡中得神托夢,始覺此乃人間福地,於是定居開墾本地。天上聖母係涂姓祖佛,廟為涂遠等開墾興建,一所名曰「開基祖廟」小廟,奉祀天上聖母,朝夕膜拜得神庇佑,家丁興旺逐漸形成大村莊,因本村落「康榔」遍野,故涂姓祖先取康榔為名,本村莊名號「大康榔」,人口達千餘戶,地處平坦,四面交通暢行,為本省開基最早村落。 先祖涂氏等為顧念村落發展迅速,實受「天上聖母」之護佑,為使眾弟子朝拜方便,乃立意募工捐集,於康熙廿九年四月擇在庄中以土石建造土廟一座,由是遠近人士悠然而來參拜繞境盛開俎豆馨香。至雍正八年涂和獻金二百兩銀,暨集其他捐金,湊足千圓、擴建廟宇原址,為涂氏宗族祖故曰「開基祖廟」村落亦由此而興,是時己繁衍一大庄社,成古時諸羅山縣大康榔西堡,為西堡之堡頭北轄至西螺,南管至蕃仔田(今隆田)附近有德化廊,安溪厝、占富厝、施寮厝、應菜埔、朴仔腳、下竹圍等村落皆屬大康榔西堡所管轄,護境安居所週知,天上聖母在大康鄉榔西堡開基,如此而形。 道光丙申年(西元一八三六年)即民前一三七年,鳩資重建廟宇於現址,奠立起稍為規模之廟宇,建材皆取之於唐山、花理石大柱、福州福杉、圓大而高、上繪故事、古時人物、飛禽走獸、花木山水、配繪八仙、栩栩如生、無不綺麗雅緻精巧出奇、亭頂嶙峋巧緻、青龍舞爪、能瞻遠近、地面黝然生光,全廟裝飾華麗光彩奪目,富麗堂皇,不愧為當時出名全台之大康榔西堡開基祖廟,香煙鼎盛民康物富,進香者絡繹不絕為本廟中興之始也,逮至民國四十四年(西元一九五五年)再倡議重建,於民國四十五年三月完工並舉行入火安座祭典移駕歸位。
